A Robust System for High-titer, Platform-agnostic AAV Production in HEK293 Cell Lines

Successful Adeno-Associated Virus (AAV) programs requires careful choosing of critical production platform components such as engineered cell lines, plasmids and transfection reagents. Amongst them, cell culture medium being the environment of all upstream AAV production steps, is foundational to productivity and efficiency of packaging. Here we present a flexible cell culture media system that simplifies process development and optimization by demonstrating broad compatibility with commercially available AAV production components. This same system supports process development strategies that yield viral titers of >1e12 vg/mL from crude lysate and seamless scale-up to a 3-L bioreactor.
