AAV producer cell lines (PCLs) offer higher and consistent productivity, better scalability and process robustness, reduced cost of goods at scale compared to transient transfection based production. However, PCL adoption is often restrained by the extended timeline of cell line construction. In this study, we present a streamlined workflow that reduces the time from DNA design to delivery of a research cell bank in five months, narrowing the timeline gap between the two production approaches. In parallel, we established standard manufacturing processes to ensure consistent product quality, including viral yield, potency and purity.
