Bone marrow is the soft tissue found in the interior of bones and is
the origin of hematopoiesis, the process in which blood cells are derived from
hematopoietic stem cells (HSC). Stem cells require specific microenvironments,
or niches, for regulation of proliferation and differentiation.
The bone marrow niche contains various types of bone marrow cells, including hematopoietic stem
and progenitor cells (HSPC), osteoblasts, mesenchymal stem cells (MSC), Chimeric Antigen
Receptor (CAR)-expressing cells, and stromal cells1.
There are two known subsets of the bone marrow niche - the osteoblast niche and the vascular
niche. While there is still debate as to the exact mechanisms, it appears as
though the osteoblast niche is responsible for self-renewal and maintenance of long-term
HSCs (LT-HSCs), while the vascular niche is responsible for the mobilization of
HSCs into the bloodstream where they differentiate into mature blood cells2.
Bone marrow cells are of increasing interest not only for their ability
to reconstitute hematopoiesis, but also for a more thorough understanding of
bone marrow niches and bone marrow cells’ specific roles in hematopoiesis, the
immune system, and human disease.
When comparing bone marrow cells to other sources of hematopoietic cells,
bone marrow cells are unique in that they provide researchers the ability to work
with large numbers of cells from a single donor or investigate differences in donors
of various ages, genders, or ethnicities.
