Various Nucleofector™ Kits and corresponding Amaxa™ Optimized Protocols are available for the transfection of rat cardiomyocytes using the different Nucleofection™ Platforms.
For the transfection of rat cardiomyocytes in the 4D-Nucleofector™ System and the 96-well Shuttle™ System we recommend using the Primary Cell Optimization Kits and the respective optimization protocols. Optimal Nucleofection™ conditions are transferable between these three systems. Rat cardiomyocyte specific kits are available for the Nucleofector™ II/2b Device.
Nucleofection™ of neonatal rat cardiomyocytes
Primary neonatal rat cardiomyocytes were transfected by Nucleofection™ with a plasmid encoding DsRed. 2 days post Nucleofection™, the cells were analyzed by fluorescence microscopy. Upper left picture shows DsRed expressing cells. Cardiomyocytes stained with FITC-labeled tropomyosin (a fibrous protein that is part of the muscle fibers) antibody are shown in the upper right picture. The lower left picture is an overlay of both upper images.

For the 4D-Nucleofector™ Kits "L" and "S" indicate the Nucleocuvette™ Vessel format. L = 100 µl single Nucleocuvette™, S = 16-well Nucleocuevtte™ Strip.
