Various Nucleofector™ Kits and corresponding Amaxa™ Optimized Protocols are available for the transfection of human dendritic cells using the different Nucleofection™ Platforms. For the transfection of human dendritic cells in the 4D-Nucleofector™ and 96-well Shuttle™ System we recommend using the Primary Cell Optimization Kits and the respective optimization protocols. Optimal Nucleofection™ Conditions are transferable between these three systems. Human dendritic cell specific kits are available for the Nucleofector™ II/2b Device.
Nucleofection™ of human dendritic cells
Human dendritic cells were differentiated from monocytes using 100ng/ml IL-4 and 50 ng/ml GM-CSF. On day 6 cells were transfected by Nucleofection™ with plasmid DNA encoding the green fluorescent protein (eGFP). 24 hours post Nucleofection™ cells were analyzed by flow cytometry (FACS).
