Sertoli cells are highly specialized and essential for maintenance of testicular homeostasis and male reproductive viability. Functions attributed to Sertoli cells include protection of the developing germ cells, secretion of factors that nurture germ cells and orchestration of temporal and spatial control of apoptosis, mitosis and meiosis during the complex process of spermatogenesis. The number of viable and productive spermatozoa is tightly correlated to the number of mature Sertoli cells. Sertoli cells form the tight junction lining of the seminiferous tubule that creates the blood-testis barrier (BTB). Similar to the blood-brain barrier, the BTB regulates the molecular exchange between the circulatory system and testis as well as protects haploid spermatid and spermatozoa from immunological destruction.
Cryopreserved Clonetics™ Sertoli Cells are shipped in the third passage. They are assured to be ≥ 70% confluence by day 12 in culture when thawed and maintained properly and express GATA-4 and Sox- 9. All cells test negative for mycoplasma, bacteria, yeast, and fungi. HIV-1, hepatitis B and hepatitis C are not detected for all donors and/or cell lots. A Certificate of Analysis is provided for each cell lot purchased.
Recommended medium: DMEM:F12 (catalog no. 12-719F or equivalent) + 5% Fetal Bovine Serum (sold separately).
To prevent bacterial contamination, 5 ml of penicillin/streptomycin (Lonza no. 17-602E or equivalent) may also be added to 500 ml of DMEM:F12 with a pipette for a final concentration of 100 units/ml potassium penicillin and 100 μg/ml streptomycin sulfate. It is not recommended to use gentamicin/amphotericin-B to prevent bacterial contamination, as these reagents have been shown to effect cell growth.
